B -- CONSTRUCT A RAINBOW TROUT NORMALIZED CDNA LIBRARY POC Lisa M. Botella, Point of Contact, Eileen LeGates, Contracting Officer, (215)233-6551, fax (215) 233-6558 The USDA, Agricultural Research Service (ARS), intends to negotiate solely with Life Technologies, Inc. in Rockville, MD to construct a large insert rainbow trout (cDNA) normalized library. The library shall be furnished and delivered (F.O.B. Destination) to the USDA, ARS, National Center for Cool and Cold Water Aquaculture at the National Conservation Training Center in Shepherdstown, West Virginia, to be used as the primary resource for obtaining gene sequence information. In addition, individual clones will be selected for physical and genetic mapping, providing comparative mapping information between rainbow trout and other species. A primary library will be constructed (directionally cloned) using RNA or tissue supplied by the Government, SuperScript II RNase H-RT, ElectroMax DH10B cells and pCMV Sport 6.0 vector. The library will be amplified 10,000 to 100,000 fold using a semi-solid agarose procedure. The library shall be normalized at COT-500 or greater using subtraction technology, and electroporated into competent E.coli to produce a library 1x 106 clones. The primary library will have greater than 3 x 106 clones. Twenty-three (23) random clones will be picked to determine that the average insert size and % vector with inserts. The average insert size shall be 1 Kb. At least twenty (20) of the twenty-three (23) clones will contain inserts. The Government shall provide the following rainbow trout tissue samples: liver, spleen kidney, gill, brain, and muscle. All remnants of the primary and amplified libraries shall be sent to the Government upon completion of the normalized library. The library producer will not retain any rights to the results of the research that utilizes the clones in the cDNA library, nor prevent the publication of these results by the Agricultural Research Service. The Government has the right to copy the library for the purposes of maintaining the library and for use in their research programs. Published research that utilizes library clones will acknowledge the contributionof the library producer. Companies with the demonstrated capability and financial capacity to provide and service such equipment must submit a written substantive statement outlining the Company's and systems' capabilities to the Contracting Officer within 15 days of publication of this notice.
