Description
DESCRIPTION: The National Institutes of Health - NICHD intends to procure the services of a contractor to provide a Conditional KO Mouse Model Generation Using CRISPR/Cas9.�� The Section on Molecular Dysmorpholgy investigates molecular, biochemical, cellular, and developmental processes that underlie genetic syndromes. Specifically, this research has focused on two rare genetic disorders, Smith-Lemli-Opitz syndrome, and Niemann-Pick Disease, type C1. Smith-Lemli-Opitz syndrome (SLOS) is an inborn error of cholesterol synthesis that results in birth defects and cognitive impairment. Niemann-Pick Disease, type C1 (NPC1) is a neurodegenerative, lysosomal storage disease due to impaired intracellular cholesterol transport. The goal of his sections research efforts is to combine both basic science and clinical expertise to develop and test novel therapeutic interventions for SLOS and NPC1. This project seeks to generate a conditional knockout of the Slc1a6 in mice. The objective of this project is to generate a floxed mouse model.� The Lab needs to use Crispr/Cas9 �to target the Slc1a6 gene in mouse embryonic stem cells generating a knockout clone. This clone will be injected into a developing blastocyst to generate a chimeric mouse that will then be used to establish this conditional knockout line. The Section on Molecular Dysmorpholgy has previously demonstrated the role Slc1a6 in Niemann-Pick Disease, type C1 (NPC1) neuroinflammation specifically within the Purkinje neurons critical from the ataxia noted as a hall mark of this disease. There is currently no Slc1a knockout mice available. As a model will be needed to further study the role of this gene in NPC1 neuroinflammation the laboratory would like to generate a cre specific model system this will serve 2 purposes. First, we will be able to specifically determine the role of this gene in the disease progression and second, we will be able to use this system specifically ribo-tag gene expression within the Purkinje neurons. The ability to look specifically at these cells has been hampered even with� advances in single cell RNAseq due to the size of the neurons. Stemcell is the only company that can meet the Lab�s needs and provide the product that will help enhance the research, using a Pronuclear Microinjection and Embryo Implantation, Genotyping� This proposed procurement is for a service for which the government intends to solicit and negotiate on a sole source basis to Applied StemCell located at� 521 Cottonwood Drive, Milpitas, CA, 95035. �In order to comply with the requirements in this Special Notice, the responses should be based on our interpretation of the general requirements provided below. Note 22. The proposed contract action is for a continuation of previous services for which the Government intends to solicit and negotiate with only one source under the authority of FAR 6.302. Interested persons may identify their interest and capability to respond to the requirement or submit quotations. This notice of intent is not a request for competitive quotes. However, all quotes received within the time and date of publication of this ""Special Note 22"" will be considered by the Government. A determination by the Government not to compete with this proposed contract based upon responses to this notice is solely within the discretion of the Government. Information received will normally be considered solely for the purpose of determining whether to conduct a competitive procurement. HOW TO RESPOND: In order to compete for this project, interested parties must be registered in the Central Contracting Registration and submit a price quotation by 10:00 am, ET, July 20th 2020.
