Description
This is a SOURCE SOUGHT NOTICE in support of the Puget Sound VA Healthcare System in Seattle, WA, for market research purposes only to determine the availability of potential businesses with capabilities to provide the services described below. Potential offerors are invited to provide feedback via e-mail to Joshua Anderson at JOSHUA.ANDERSON2@VA.GOV. Responses will be used to determine the appropriate acquisition strategy for a potential future acquisition. All responses due by September 19th, at 1300. The Capabilities Statement for this sources sought is not expected to be a Request for Quotations, Request for Proposals or Invitation for Bids, nor does it restrict the Government to an ultimate acquisition approach, but rather the Government is requesting a short statement regarding the company s ability to provide the services outlined in the draft SOW below. Any commercial brochures or currently existing marketing material may also be submitted with the capabilities statement. Submission of capabilities statement will assist our office in tailoring the requirement to be consistent with industry standards. The capabilities will be evaluated solely for the purpose of determining to Set-Aside for the Small Business (SB) Community or to conduct as an Unrestricted Procurement. Other than small businesses may respond to this notice in the event the market does not indicate SB interest. This synopsis is for information and planning purposes only and is not to be construed as a commitment by the Government. The Government will not pay for information solicited. Respondents will not be notified of the results of the evaluation. Note: Do not include Proprietary, classified, confidential, or sensitive information in responses. Not responding to this Sources Sought does not preclude participation in any future or potential solicitation, or Request for Quotation. It is the intent of the Puget Sound VA Medical Center to use the information gathered for market research purposes only. If a formal solicitation is released, it will be posted via the Federal Business Opportunity (http://www.fbo.gov). DESCRIPTION OF THE REQUIREMENT: STATEMENT OF WORK (SOW) 1. Contract Title. Pronuclear DNA Transgenic Mice 2. Background. Research investigator Dr. Brian Kramer requires pronuclear DNA micro-injection of mouse embryos for making transgenic mice as part of his Alzheimer s research at VAPSHCS. Dr. Kraemer will provide a plasmid construct to the vendor for microinjection into mouse embryos. The plasmid will contain all the DNA sequences necessary for the expression of any desired protein of interest, including promoter sequences and protein coding sequences. The DNA will be purified. The proteins of interest will include those involved in the Unfolded Protein Response. Future research will use transgenic mouse models that overexpress these proteins to determine whether this strategy can reduce pathology in an intact mammalian brain, to set the stage for future possible clinical development in human patients. 3. Scope. The transgenic mouse service provider must: Perform pronuclear DNA microinjections into single cell 0.5 day old embryos to generate transgenic pups. Dr. Kraemer will send a plasmid construct of recombinant DNA molecules for microinjection into the 0.5 day embryos to the service provider, where they will use microinjection techniques to insert the DNA into the single cell mouse embryos. This technique requires highly skilled technicians, and only a few centers in the country produce a high rate of success with these methods. Be able to inject approximately 100 embryos per injection day. Use the mouse strain C57BL/6, which are the strain of mice used in Dr. Kraemer s other projects. Be able to demonstrate that they can obtain a minimum of 10-15% transgenic pups out of the total number of pups born following pronuclear injection. House the embryo recipient female mice and any pups born until they are approximately 3 weeks of age, at which time a tail tip sample will be taken from all pups. Use a FedEx number provided by Dr. Kraemer to ship tail samples for screening of the transgene. Tail samples will be tested by PCR and/or DNA sequencing to determine which ones contain the desired transgenic modification. Continue to house any pups identified as potentially transgenic for a period of 2-3 weeks before shipping to VAPSHCS Animal Research Facility. After positive pups are identified, ship the positive pups to Dr. Kraemer (see 9. h.). Provide veterinary care to all mice (mothers and pups) as needed throughout the pregnancy and subsequent period until successful genotyping and up to shipment. Be located at an AAALAC accredited animal facility, to ensure a high level of care for the animals. Be flexible and able to set up for as many as 12 days injection services but allow for the possibility of less that 12. Be able to set up the contract to accommodate the possibility for up to 12 injections as something like a cost-reimbursement contract where VA pays for allowable incurred costs only. 4. Specific Tasks. Outline of pronuclear DNA microinjection: a. Microinjection using the provided samples will be scheduled once the service paperwork has been approved by the directors of this facility. b. Mice generated from the pronuclear DNA microinjections will be housed under the Transgenic Mouse Shared Resource protocol number until shipping to Dr. Kraemer. c. At three weeks of age, resulting pups will be weaned, tailed, and ear punched. A dam from each litter will be used for serology purposes. d. Dr. Brian Kraemer will provide a FedEx number to use for shipping tail samples to be screened. e. Positive pups will be identified by the investigator; a transfer request will be submitted for shipment of the positive mice to the Veteran s Affairs Puget Sound Health Care System Animal Research Facility. f. Pending approval of serology screening results by the Vanderbilt Department of Animal Care and the receiving institution, the transgenic pups will be shipped to the Veteran Affairs Puget Sound Health Care System for breeding. 5. Performance Monitoring The performance of the contractor is supervised by the Principal Investigator, Dr. Brian Kraemer, who will do genetic testing of the pups prior to their shipment to VA. Dr. Kraemer will provide for shipping costs to have tail snips sent to VA. 6. Other Pertinent Information or Special Considerations. a. This work is for projects approved though the VAPSHCS Research and Development Committee on protocols RDIS0007/MIRB0336. b. The vendor must have a PHS Assurance Number assigned and operate under the guideline provided by the Public Health Service Animal Welfare Assurance Statement. (see attached email PDF file: Vanderbilt PHS Assurance _ RE_ Pronuclear DNA email for preferred vendors statements regarding PHS Assurance) c. Use of this type of transgenic mouse has been approved by the VAPSHCS Institutional Animal Care and Use Committee (IACUC) on protocol # 0765. Arrangements will be made in accordance with VAPSHCS animal research facility (ARF) standards and the service provider s facility for the proper exchange of pathogen free certificates and any other necessary certifications needed by VAPSHCS ARF prior to shipment. Approval for each animal shipment (up to 4) will be obtained from VAPSHCS ARF prior to each shipment being made. d. Packaging, Packing and Shipping Instructions. Animals must be shipped in a manner that retains their health and viability. Animals must arrive at VAPSHCS alive and with normal vital signs: weight, alertness, appetite and exhibiting normal physical and social behaviors. Animals must be genetically identical to those tested using tail snips sent to the VAPSHCS Kraemer Lab prior to their arrival. Animals must comply with VAPSHCS ARF pathogen free certification status.
