Description
THIS IS A NOTICE OF INTENT TO AWARD A SOLE SOURCE ORDER AND NOT A REQUEST FOR QUOTATIONS. The National Institute of Environmental Health Sciences (NIEHS), Research Triangle Park, NC intends to negotiate on a sole source basis and issue an order with University of California, Irvine, CA 92697 under the authority of Federal Acquisition Regulation (FAR) 13.106-1(b). This request is for 20 chemicals to be tested in agonist mode on 4 nuclear receptors (RXRa, PPARy, RARa, and GR, plus 1 extra control for GR) during the base year. There will be 2 option years for testing of an additional 5 chemicals each option year. The Blumberg Laboratory at the University of California will test the ability of certain compounds identified in bioinformatic analysis of high throughput screening (HTS) experiments performed by the NTP (Tox21) to activate nuclear hormone receptors relevant to adipogenesis (agonist, aka receptor activation assays). The receptors contemplated to be tested under this contract include the following: human peroxisome proliferator activated receptor gamma (PPARγ); human 9-cis-retinoic acid receptor alpha (RXRα); human glucocorticoid receptor (GR); and human retinoic acid receptor alpha (RARα). Chemicals shall be tested together with appropriate negative controls (tk-MH1004-luc and MMTV-luc reporters plus empty effector plasmids). Compounds to be tested in agonist assays will be evaluated in triplicate at 7 concentrations between 10-5 - 10-11 M. The test compounds will be supplied dissolved in DMSO to make 1000x stock solutions before adding to media. All assays are done in triplicate maintaining a final DMSO concentration of ≤ 0.05% v/v. The experimental endpoints of agonist assays will be evaluated as relative light units normalized to β-galactosidase transfection controls and compared with solvent controls (Janesick et al, 2015c). Positive control chemicals for each receptor will be tested at 7 concentrations between 10-5 - 10-11 M as follows: PPARγ - rosiglitazone, RARα - TTNPB or 9-cis retinoic acid (RA), RXRα - AGN194204 or 9-cis RA, GR - dexamethasone. Dose response curves of test chemicals will be compared with those of control chemicals for each receptor tested. Non-linear regression analysis will be performed in GraphPad Prism 5.0 to determine EC50 values (or IC50 values if added to the scope of the experimental design) for all active chemicals. Basic statistical analysis will be conducted in GraphPad Prism 5.0 using one-way ANOVA to determine differences in fold induction (or reduction) among treatment doses and negative control (0.05% DMSO). This will be followed by a Dunnett's post-hoc test to ascertain statistical significance for each dose compared to control (DMSO). Further statistical analysis and curve modeling will be performed by NTP personnel. The laboratory will provide NTP with a final report containing a detailed experimental approach, the raw data and normalized data that determine EC50 values for each of the test compounds with respect to receptor activation. Award is subject to approval of the Sole Source Justification. This notice of intent is not a request for competitive proposals or quotations; however, an interested party may submit a statement of capabilities if they believe they can meet the above requirement. The statement of capabilities must be submitted in writing and contain sufficient detail to allow the Government to determine if the party can provide the requirement. A determination by the Government not to compete this proposed action based upon responses to this notice is solely with the discretion of the Government. All responses must be received within 15 days from the date of publication of this synopsis. Information received will normally be considered for the purpose of determining whether to conduct a competitive procurement. If no affirmative responses are received within 15 days of this notice, an award will be made to University of California. The NAICS Code/Size is 541990/$15.0 Million. This procurement is to be processed using Simplified Acquisition Procedures. There is no solicitation package available. No faxed documents will be accepted.
