Description
This Sources Sought Notice (SS) is for information and planning purposes only and shall not be construed as a solicitation or as an obligation on the part of the National Heart, Lung, and Blood Institute (NHLBI). The purpose of this SS is to identify businesses that are interested in Exome Genotyping of 3,360 DNA samples from the USA, Africa and China. The NHLBI does not intend to award a contract on the basis of responses nor otherwise pay for the preparation of any information submitted. As a result of this Sources Sought notice, the NHLBI may issue a Request for Quote (RFQ). THERE IS NO SOLICITATION AVAILABLE AT THIS TIME. However, should such a requirement materialize, no basis for claims against NHLBI shall arise as a result of a response to this Sources Sought notice or the NHLBI's use of such information as either part of our evaluation process or in developing specifications for any subsequent requirement. The NHLBI is seeking capability statements from all eligible businesses, under the North American Industry Classification System (NAICS) code 541711, Research and Development in Biotechnology, Size Standard in number of employees: 500. Background: The mission of the National Human Genome Research Institute (NHGRI), Center for Research on Genomics and Global Health (CRGGH) is to advance research into the role of culture, lifestyle, genetics and genomics in health disparities. The CRGGH lab has ongoing large-scale genetic epidemiology studies of metabolic disorders including obesity, type 2 diabetes (T2D), and hypertension, dyslipidemia in African Americans, Africans and Chinese. Over 8,000 DNA and other biological specimen have been collected, processed and stored on study participants. The goal of these projects is to identify the genetic and non-genetic contributors to several metabolic disorders with the ultimate goal of informing the health disparity observed in African Americans compared to their European counterparts. By developing genetic epidemiology models to investigate the complex interaction between genetic and lifestyle factors, CRGGH hopes to shed light on the patho-biology of metabolic diseases in different human populations especially African Americans. Objectives for the project are: 1. Use whole exome array technology to genotype 3,360 DNA samples. 2. Conduct whole-exome association study on several traits including obesity, T2D and related complications, blood pressure parameters (hypertension, systolic and diastolic BP), lipid parameters (HDL, LDL, triglycerides and total cholesterol) and kidney function parameters (eGFR, creatinine, and Albumin). 3. Perform a replication study of whole-exome association study findings in independent samples of African Americans, Africans, Chinese and European ancestry populations. Contractor Requirements A. The Contractor shall use whole exome array technology to genotype 3,360 DNA samples. Provide analysis of 1,056 DNA samples collected and located in Suizhou, China. Provide analysis of 2,304 DNA samples collected and located in Bethesda, Maryland. The Contractor's exome array technolgy shall the following specifications: Feature Specification Validation method 318,983 wet-lab genotyped markers Total validated markers 318,983 Non synonymous coding SNPs 247,546 Indels 35,137 Chromosome X/Y 6,918/161 Identity by descent SNPs 5,478 GWAS tag markers 5,053 Ancestry informative markers 4,507 Synonymous SNPs 4,367 HLA region markers 2,262 ESP SNPs 861 SNPs for DNA fingerprinting 263 Mitochondrial DNA/miRNA 207/250 Customizable markers <113,000 Exome genotyping will quarrying more than 300,000 coding SNPs including non-synonymous and synonymous SNPs as well as variants in splice and stop codons. Content of the array will also include over 20,000 single-base, complex indels and AIMs - Ancestry informative markers to measure Latino and African American ancestry. To facilitate merging exome data with genome-wide association study datasets, it is expected that the data will include thousands of SNPs (> 5,000) reported in the NHGRI Catalog of Published Associations and other sources. Exome array will also generate data on human leukocyte antigen (HLA) genes, chromosome Y, mitochondrial DNA, and microRNA sequences. Finally, the data will be customized with additional SNPs (up to 100,000) from CRGGH own candidate genes. B. Upon receipt, samples are inspected and logged in against the government provided file. Discrepancies, if any, are rectified and a project specific database is established. For sample tracking, labels are printed with the government provided sample ID as well as the contractor assigned sample sequential processing ID. Labels are attached to all processing tubes. The generated data and leftover DNA samples are to be shipped to CRGGH by the contractor. Government Responsibilities A. The Government/CRGGH will provide the contractor the following items: 1. All required barcode labels for the project; 2. Develop data entry screen; 3. Develop and implement quality control procedures for the project; and 4. Provide timely feedback on materials received from contractor. B. The Government/CRGGH willshipthe 2,304 located at NIH to the contractor; the Gonverment/CRGGH will also provide the contractor with the samples' manifest as an electronic file (i.e., Microsoft Excel worksheets). C. Samples to be received by the contractor in IATA650 compliant shipping materials. Reporting Requirements and Deliverables A final lab report to the Governent will include a description of completed tasks and generated data will be prepared by the contractor. Interested parties are expected to review this notice to familiarize yourself with the requirements of this project; failure to do so will be at your firm's own risk. Interested parties should submit a tailored capability statement for this requirement, not to exceed 20 single-sided pages (including all attachments, resumes, charts, etc.) presented in single-space and using a 12-point font size minimum, that clearly details the ability to perform the aspects of the notice described above and in the attached draft statement of work. All proprietary information should be marked as such. Statements should also include an indication of current certified business status; this indication should be clearly marked on the first page of your capability statement (preferably placed under the eligible small business concern's name and address). Responses will be reviewed only by NIH personnel and will be held in a confidential manner. All capability Statements sent in response to this SOURCES SOUGHT notice must be submitted electronically (via email) to James Richards, Acting Contract Specialist, at richardsje2@mail.nih.gov in either MS Word or Adobe Portable Document Format (PDF), May 9, 2012, 7:30 AM, EST. All responses must be received by the specified due date and time in order to be considered.
