Description
The United States Department of Agriculture (USDA), Agriculture Research Service is issuing the following requirement as a Combined Synopsis/Solicitation: Vendor shall produce 12 grams of endotoxin free Lysostaphin-TAT-His(6) (Lyso-TAT) a fusion of lysostaphin to the HIV Trans Activator of Transcription (TAT) protein transduction domain with a 6 x His Tag for nickel column purification. The fusion protein (270 residues) is encoded on the pET21a (Novagen) IPTG inducible plasmid. USDA has a protocol using NiNTA nickel column matrix (Qiagen) that yields 2 mg per liter of shaker flask grown E. coli harboring the Lyso-TAT vector. Prior to awarding this requirement, the vendor shall include a demonstrated purification on a small volume (e.g. 10 liter) of fermentation grown E. coli BL21 (DE3) to demonstrate that the target amount (12 g) can be produced at the concentration and quality anticipated for the full scale grow up. The final end-product is required by April 15, 2012. USDA shall send the Lyso-TAT encoding plasmid and purification protocol to interested parties. The final product shall be demonstrated via SDS PAGE and OD(280) to be >98% pure., at a connection of >10 mg/ml Lyso-TAT, endotoxin free, in: 50Mm NaH2P04, 300Mm NaCl; 2500mM imidazole; 30% glycerol and pH8.0. he anticipated award date is March 19, 2012. This procurement is reserved exclusively for small businesses. This notice is for informational purposes only and is not to be construed as a commitment by the Government. Potential sources who believe they can provide the services listed above are invited to submit substantiating documentation in writing to the identified point of contact. Information received will be considered solely for the purpose of determining whether to conduct a competitive procurement. Oral communications are not acceptable in response to this notice. A determination by the Government to compete this proposed contract based on responses to this notice is solely at the discretion of the Government
