Sources Sought Notice No.: NHLBI-CSB-(AR)-2011-251-CRS Title: Instrument That Allows Measurement of mRNA With High Sensitivity This Sources Sought Notice (SS) is for information and planning purposes only and shall not be construed as a solicitation or as an obligation on the part of the National Heart, Lung, and Blood Institute (NHLBI). The purpose of this SS is to identify qualified small business concerns [including Small Disadvantaged Businesses (SDB), Woman-owned Small Businesses (WOSB), Historically Underutilized Business Zone (HUBZone) Small Businesses, Veteran-Owned Small Businesses (VOSB) and Service-Disabled Veteran-owned Small Businesses (SDVOSB] that are interested in and capable of providing the instrument that allows measurement of mRNA With High Sensitivity. The NHLBI does not intend to award a contract on the basis of responses nor otherwise pay for the preparation of any information submitted. As a result of this Sources Sought notice, the NHLBI may issue a Request for Quote (RFQ). THERE IS NO SOLICITATION AVAILABLE AT THIS TIME. However, should such a requirement materialize, no basis for claims against NHLBI shall arise as a result of a response to this Sources Sought notice or the NHLBI's use of such information as either part of our evaluation process or in developing specifications for any subsequent requirement. The NHLBI is seeking capability statements from all eligible small business concerns [particularly Small Disadvantaged Businesses (SDB), Woman-owned Small Businesses (WOSB), Historically Underutilized Business Zone (HUBZone) Small Businesses, Veteran-Owned Small Businesses (VOSB) and Service-Disabled Veteran-owned Small Businesses (SDVOSB)] under the North American Industry Classification System (NAICS) code 541990 with a size standard of $7.0 Million. Based on the responses received from this SOURCES SOUGHT announcement, the proposed acquisition may be solicited as a Total Small Business Set-Aside. All eligible small business concerns responding to this Sources Sought Announcement must have the capabilities to provide the below services upon receipt of order. Interested parties are expected to review this notice to familiarize yourself with the requirements of this project; failure to do so will be at your firm's own risk. Background The Translational Immunology Section at National Institute of Arthritis and Musculoskeletal and Skin Diseases (NIAMS), is critically involved in several studies which requires assessment of the expression levels of genes from cohorts of clinical patients. In order to properly support studies on the genetic basis of autoimmune disease- NIAMS requires technologies that allow high throughput measurement of transcript abundance. This could be achieved via different technologies but, in particular, NIAMS needs an instrument that allows measurement of mRNA with high sensitivity (possible as low as 1 copy of mRNA per cell) and that will be important to be able to assess transcript abundance directly without any amplification step and with a digital readout output. Specifications shall include: Level of Multiplexing -Up to 550 Gene Targets per Assay Recommended Amount of Starting Material- 100ng of total RNA, or lysate from ~10,000 cells Sample Types Supported- Total RNA, Cell Lysates in GITC, FFPE derived total RNA and PAXgene lysed whole blood Reaction Volume- 30 μL Limit of Detection- 0.5fM spike-in control (~1 copy per cell); 90% of the time Fold Change Sensitivity- >1.5 fold (>5 copies per cell)>2 fold change >(1 copy per cell) Linear -Dynamic Range 7 X 105 Total Counts Throughput -12 samples <2.5 hours Digital Analyzer Throughput- 12 samples/4 hours (up to 72 samples per day Disclaimer and Important Notes: This notice does not obligate the Government to award a contract or otherwise pay for the information provided in response. After a review of the responses received, a pre-solicitation synopsis and solicitation may be published in Federal Business Opportunities. However, responses to this notice will not be considered adequate responses to a solicitation. Confidentiality. The Government reserves the right to use any non-proprietary technical information in any resultant solicitation(s).
