Description
This is a combined synopsis/solicitation for commercial items prepared in accordance with the format in Subpart 12.6, as supplemented with additional information included in this notice. This announcement constitutes the only solicitation; proposals are being requested and a written solicitation will not be issued. The brains (mice, rat and guinea pig) should not be cut into two hemispheres. In order to obtain similar coordinates, blocking and embedding for both paraffin and frozen sectioning have to be precise. The total number of slides needed for H&E, Thionin, TUNEL, FJ-B and immunohistochemistry are described below. 1. Total number of paraffin blocks, slides and H&E stained slides generated from each species. (a)50 mouse brains: 150 paraffin blocks (3 different sites/coordinates/levels X 50 brains), 600 slides (4 slides per block) and 150 H&E stained slides (1 slide per block) will be generated. (b)50 mouse brains: 150 paraffin blocks (3 different sites/coordinates/levels X 50 brains), 600 slides (4 slides per block) and 150 H&E stained slides (1 slide per block) will be generated. (c)90 guinea pig brains: 270 paraffin blocks (3 different sites/coordinates/levels X 90 brains), 1080 slides (4 slides per block) and 270 H&E slides (1 slide per block) will be generated. (d)75 primate hemi brains: 225 paraffin blocks (3 different sites/coordinates/levels X 75 brains), 900 slides (4 slides per blocks) and 225 H&E slides (1 slide per block) will be generated. Special requirements: positive-charged slides should be used, no exceptions. For mice and rat brains, 3 sections per slide are required. For guinea pig and primate hemi brains, 2 sections per slides are required. All unstained slides (3 slides per block per species) are to be returned to the client for further processing. 2. Total number of cryoblocks, cryosections and stained slides (Thionin) (a)50 mouse brains: 100 cryoblocks (2 different sites/coordinates/levels X 50 brains) and 200 thionin stained slides (2 slides per block) will be generated. (b)50 mouse brains: 100 cryoblocks (2 different sites/coordinates/levels X 50 brains) and 200 thionin stained slides (2 slides per block) will be generated. (c)90 guinea pig brains: 180 cryoblocks (2 different sites/coordinates/levels X 90 brains) and 360 thionin stained slides (2 slides per block) will be generated. (d)75 primate hemi brains: 150 cryoblocks (2 different sites/coordinates/levels X 75 brains) and 300 thionin stained slides (2 slides per block) will be generated. Special requirements: All sections must be mounted on gelatin-coated slides. All cryoblocks must be cut at 30 microns and through. Every section must be collected and stored in cryoprotective buffer and return to the client. 3. TUNEL will be conducted on paraffin sections. (a)25 mouse brains X 3 sites/coordinate/levels X 1 slides per site = 75 slides (3 sections per slide) (b)25 rat brains X 3 sites/coordinate/levels X 1 slides per site = 75 slides (3 sections per slide) (c)60 guinea pig brains X 3 sites/coordinate/levels X 1 slides per site = 180 slides (2 sections per slide) (d)50 primate hemi brains X 2 sites/coordinate/levels X 1 slides per site = 100 slides (2 sections per slide) Only certain brains from each species will be stained with TUNEL. 4. Fluoro-Jade B (FJ-B) will be conducted on frozen serial sections (a)50 mouse brains X 2 sites/coordinate/levels X 2 slides per site = 200 FJ-B slides (3 sections per slide) (b)50 rat brains X 2 sites/coordinate/levels X 2 slides per site = 200 FJ-B slides (3 sections per slide) (c)90 guinea pig brains X 2 sites/coordinate/levels X 2 slides per site = 360 FJ-B slides (2 sections per slide) (d)75 primate hemi brains X 2 sites/coordinate/levels X 2 slides per site = 300 FJ-B slides (2 sections per slide) 5. Immunohistochemistry will be conducted on frozen serial sections. (a)50 mouse brains X 2 sites/coordinate/levels X 6 sections per site = 600 sections (b)50 rat brains X 2 sites/coordinate/levels X 6 sections per site = 600 sections (c)90 guinea pig brains X 2 sites/coordinate/levels X 6 sections per site = 1080 sections (d)75 primate hemi brains X 2 sites/coordinate/levels X 4 sections per site = 600 sections
