Description

Description The National Cancer Institute (NCI), Division of Cancer Epidemiology & Genetics (DCEG), Occupational and Environmental Epidemiology Branch (OEB) plans to procure on a sole source basis teleomere length and mDNA copy number analysis from University of Milan & IRCCS Maggiore Hospital, Mangiagalli and Regina Elena Foundation, Via San Barnaba 8, 20122 Milan, Italy. This acquisition will be processed under FAR Part 12 - Acquisition for Commercial Items and will be made pursuant to the authority in FAR 13.106-1(b)(2) and 13.501-(a)(1) using simplified acquisition procedures for commercial acquisitions. The North American Industry Classification System code is 541380 and the business size standard is $12.0 million. Only one award will be made as a result of this solicitation. This will be awarded as a firm fixed price type contract. Period of Performance: Twelve months from Date of award Pesticide exposure has been associated with increased cancer risk in epidemiological studies, but an understanding of potential mechanisms is lacking. There is some evidence, mostly in animal or in vitro studies, that OPs and certain herbicides may induce lipid peroxidation and/or significant changes in antioxidant enzymes, resulting in excess generation of reactive oxygen species or free radicals. It is hypothesized that these classes of pesticides may be linked to cancer via oxidative stress mechanisms; however data in humans is limited. MtDNA is a suitable biomarker of oxidative stress since it is more susceptible to reactive oxygen species than nuclear DNA. Higher mtDNA copy number has been linked to oxidative stress due to occupational exposures, such as benzene and N,N-dimethylformamide, as well as with excess cancer risk, including lung and non-Hodgkin lymphoma. Telomere shortening has been has also been linked to oxidative stress, including exposure to certain pesticides. The Agricultural Health Study (AHS) Cohort provides the unique opportunity to assess an exposure-response association between lifetime pesticide exposure on mtDNA copy number and telomere length while adjusting for important potential confounding factors. This study has the potential to provide important insights into the mechanisms by which pesticides may be linked to cancer. The contractor for this procurement shall conduct telomere length and mitochondrial DNA copy number in a sample (n=685) of Agricultural Health Study participants using DNA that has already been extracted from whole blood using the DNAQuikTM method. The contractor shall conduct telomere length analysis using real-time quantitative PCR (qPCR) in 384-well plate and mtDNA copy number using fluorescence-based quantitative PCR (qPCR) in 384-well plates. Appropriate quality control procedures for each assay will be conducted. The proposed contactor, Center of Molecular and Genetic Epidemiology at the University of Milan, has extensive experience and specializes in conducting molecular analyses such as telomere length assays for environmental epidemiological studies. This lab has conducted previous molecular analyses using AHS samples (PUR261200800172, PUR26100700252), and has demonstrated excellent CVs for telomere length assays. For consistency of laboratory methods and comparability of results, we propose the designated contractor. This is not a solicitation for competitive quotations. However, if any interested party believes they can meet the above requirement, they may submit a statement of capabilities. All information furnished must be in writing and must contain sufficient detail to allow the NCI to determine if it can meet the above unique specifications described herein. An original and one copy of the capability statement must be received at the NCI Office of Acquisitions on or before 11:00 AM EST on September 16, 2010. No electronic capability statements will be accepted (i.e. email or fax), an original and one copy must be sent to the NCI Office of Acquisitions to the address stated above.
