SF-18. Please return this form with your quote. COMBINED SYNOPSIS/SOLICITATION SUPPORT NOAA RESEARCH BY OTOLITCH MICROCHEMISRY ANALYSIS AND BY SCALE ANALYSIS IN ACCORDANCE WITH THE STATEMENT OF WORK. (I) This is a combined synopsis/solicitation for commercial items prepared in accordance with the format in FAR Subpart 12.6, as supplemented with additional information included in this notice and in accordance with the simplified acquisition procedures authorized in FAR Part 13. This announcement constitutes the only solicitation; proposals are being requested and a written solicitation will not be issued. (II) This solicitation is issued as a request for quotation (RFQ). Submit written quotes on RFQ Number WRC-10-19736 (III) The solicitation document and incorporated provisions and clauses are those in effect through Federal Acquisition Circular 2005-10. (IV) This solicitation is unrestricted and open to all vendors. The associated NAICS code is 541712. (V) This combined solicitation/synopsis is for purchase of the following commercial item(s)/services: See Attached. (VI) Description of requirements is as follows: Statement of Work: WRD-10-19736 Statement of Work for Otolith and Scale Analysis Task 1: Otolith Analyses Several investigators have documented the feasibility of distinguishing between juvenile salmonid progeny spawned by anadromous or resident maternal parents using strontium levels recorded in their embryonic otoliths (Kalish, 1990; Rieman et al., 1994, Volk et al., 2000). In salmonids, otoliths begin forming in embryos about the time a weak eye is evident, and the chemical composition of the otolith core is largely dependent upon the chemical make up of the yolk material. Since anadromous female salmon deposit yolk material in strontium rich seawater, and strontium easily substitutes for calcium in the otolith aragonite lattice, embryonic core otolith regions also show relatively high strontium levels. In contrast, assuming that the freshwater system does not have an unusually high strontium concentration, core otolith regions of resident female progeny generally have much lower strontium levels. The object of this work is to measure the Sr/Ca ratio from hatchery-reared Duckabush River steelhead and compare them with natural-origin Onchorhynchus mykiss parr collected from the same population in previous years. The steelhead to be analyzed were collected as eyed embryos from gravel redds and their egg size indicated they were produced by anadromous mothers which would have a high Sr/Ca ratio compared to eggs produced by resident (non-anadromous) females. The contractor will cast, section and prepare for analysis otoliths from the 20 steelhead, which will be analyzed for Sr/Ca ratios using Laser Ablation Inductively coupled Plasma Mass Spectrometry (LA-ICP-MS). The otoliths will be provided to the contractor by 1 September 2010. The otoliths from these smolts will be analyzed by the methods described in Volk et al. (2000). For each specimen, transects with be conducted near the primordia and at the otolith edge to represent the maternal and freshwater signals respectively. Elevated primordia Sr/Ca values will be taken as evidence for maternal anadromy, while low values indicate resident parentage. Data will be transformed to Sr/Ca atomic ratios to make classifications of anadromous or resident maternal parentage. Results will be summarized and presented in excel spreadsheets and/or Power Point Slides and Sigmaplot graphics. The contractor will compare the Sr/Ca levels in core and edge regions to Sr/Ca ratio data collected from 90 natural parr in the Duckabush River from 2006-2008 (provided by NOAA). A technical report will be produced that will determine which of the 90 parr have Sr/Ca ratios consistent with the Sr/Ca ratios in the hatchery-reared Duckabush River fish analyzed as part of this study. Anticipated Completion Date: June 1, 2011 Task 2: Scale analyses Scales collected from O. mykiss sampled from seven Hood Canal streams will be analyzed to determine age. NOAA will provide up to 1,500 scales collected from O. mykiss from various outmigration traps collected during the summer of 2010 and spring of 2011. Scales will be provided mounted on standard scale cards. Scale ages will be reported based on winter annulus formation in both fresh and saltwater. For example an adult spending two winters in freshwater and one full winter (two summers) in seawater would be recorded as 2.1+. The ‘+' refers to rapid spring/summer growth.
