This acquisition will be processed under FAR Part 12 - Acquisition for Commercial Items and will be made pursuant to the authority in FAR 13.106-1(b)(2) and 13.501-(a)(1) using simplified acquisition procedures for commercial acquisitions. Only one award will be made as a result of this solicitation. This will be awarded as a firm fixed price type contract. The Clinical Pharmacology Program (CPP) performs pharmacokinetic analysis of patient samples in support of numerous clinical studies of anticancer agents performed at the NCI. This work requires state-of-the-art instruments for separation (chromatography) and detection (mass spectrometry). CPP currently requires two UPLC-MS-MS instruments and the necessary components to replace existing and outdated LC-MS technology in order to improve sensitivity, efficiency and productivity. These components, installation and training are necessary for use with the UPLC/MS/MS systems (AB SCIEX QTRAP 5500 Mass Spectrophotometers coupled to ACQUITY UPLC) which are used for quantification of anti-cancer drugs in clinical samples, for pharmacokinetic evaluation. Pharmacokinetic studies are an integral part of almost all NCI clinical trials of new anticancer agents. The Clinical Pharmacology Program is responsible for providing this service to all CCR investigators. The system will replace existing instrumentation that is over a decade old and cannot provide the necessary sensitivity or throughput. The instrumentation and associated computer systems are required to have a range of features only available from the proposed contractor. The AB SCIEX QTRAP® 5500 System is the only mass spectrometer that offers high sensitivity, bench top triple quadrupole mass spectrometer performance and Linear Ion Trap performance in a single instrument. This instrument provides an uncompromised combination of Linear Accelerator trap and triple quadrupole functionality, uniquely allowing both qualitative and quantitative analyses to be carried out in a single experiment. New eQ TM electronics and the new Qurved LINAC® collision cell provide unmatched performance for multi-analyte analysis and fast, high resolution chromatography. The Applied Biosystems Software and Operating System are unique in that the software provides automatic, dynamic adjustment of the acquiring MRM transitions such that only compounds that elute within a specified time window are monitored. In addition, the process of creating the acquisition method to utilize this feature accepts a simple list of compound names, MRM transitions, and expected retention times and uses this list to dynamically schedule MRM scans during the experiment. (Scheduled MRM TM Algorithm). Discrete time segments are not required for scheduled MRM. The software has complete automated quantitative data processing and reporting capabilities. The software is capable of direct data transfer to popular word processing programs such as MS Word, Excel, Power Point, etc. The Applied Biosystems automated metabolite idenfication software automatically builds acquisition methods to target user defined panels of drug metabolites and automatically quantitates and facilitates the structural elucidation of these compounds through integration with ACD/MS Processor software. This automated solution greatly streamlines the difficult and time-consuming task of structural elucidation of metabolites. Finally the software is fully automated including customized data processing utilizing scripting. UPLC inlet to the AB Sciex QTrap5500 must offer rapid separation speed, high sensitivity, accuracy and the following specifications met only by the ACQUITY UPLC system. The UPLC system is required to be compatible with the AB Sciex QTrap5500 and can be directly controlled from within QTrap Analyst software. For different applications, the solvent delivery system needs to have a flow rate range of 0.010-2.0 ml/min, settable in 0.001 ml increments. The pump can deliver maximum of 2 ml/min during an analysis and up to 10 ml/min when priming. In order to achieve better separation, the solvent delivery system can deliver gradient with eleven gradient curve shapes (linear, 2 step, 4 convex, 4 concave). The Sample Manager can support up to three injections modes : full loop, partial loop and partial loop using needle overfill (PLNO). With full injector loop mode, the sample delivery precision should be within 0.3% RSD and in PLNO mode 0.2 µL injection with 1% accuracy. The system should support eCord technology which documents the methods and usage of ACQUITY UPLC columns, up to 50 sample sets, the minimum and maximum pressure and temperature, sample and injection totals for the column can be stored and accessed using ACQUITY UPLC system Console software.
