Description
THIS IS A NOTICE OF INTENT, NOT A REQUEST FOR PROPOSALS. A SOLICITATION DOCUMENT WILL NOT BE ISSUED AND PROPOSALS WILL NOT BE REQUESTED. It is the intent of the National Heart, Lung, and Blood Institute (NHLBI), for the National Human Genome Research Institute (NHGRI), to negotiate/award a purchase order to Beckman Coulter, 200 South Kraemer Blvd., Brea, CA 92821, for the purchase of a SPRIworks Library System. Beckman Coulter’s SPRIworks Fragment Library System I is a fully automated system for the preparation of library samples for the Illumina Genome Analyzer. The SPRIworks Fragment Library System method has three components: SPRI-TE Nucleic Acid Extractor instrument, method card that controls liquid handling, and a sealed cartridge prefilled with all the reagents required to prepare a single library. In addition to the necessary enzymes and buffers for the preparation of a library, the cartridge reagents include an optimized size selection capability based on SPRI chemistry. SPRI is an acronym for Solid Phase Reversible Immobilization and are covered under US Patents 5,705,628, 5,898,071 and 6,534,262. SPRI chemistry is a novel chemistry in which nucleic acids are immobilized around the surface of the particles by controlling environmental reaction conditions such as pH, salts concentration and hydophobicity and hydrophilicity. NISC (NIH Intramural Sequencing Center) is a centralized research facility for performing high-throughput, large-scale DNA sequencing and sequencing analysis. The Beckman SPRIworks Fragment Library System I would enable NISC to reduce the extensive manual labor required in library preparation for the Illumina Genome Analyzers so the personnel can focus on other preparation work. This equipment will reduce cost in additional personnel, reduce errors from manual processing since the machine is fully automated and the reagents are pre-dispensed, and provide reproducible and consistent library output. No other company has this type of instrumentation or carry these reagents. Beckman Coulter is the only company that can provide or support the reagents and equipment. SPRIworks Fragment Library Systems Overview The SPRIworks Fragment Library System has three components; The SPRI-TE Nucleic Acid Extractor, a method card that controls liquid handling and a sealed cartridge prefilled with all the reagents required to prepare a single library. SPRIworks Systems and Reagents Sole Source Specifications: 1. The vendor must provide, minimize and disclose all dangerous chemicals properties of the reagent system. SPRI-Based reagents require no need for use of chaotropic salts, which are known and classified by OSHA as corrosive chemicals. 2. The vendor system should be a fully automated library construction solution for the preparation of libraries compatible with the Illumina Genome analyzer and the Roche GS FLX DNA Sequencer. 3. The system, reagents and plastic consumables should be supplied and supported by a single vendor. 4. The vendor’s system must be capable of performing size selection with SPRI chemistry 5. The vendor’s reagent cartridge must have SPRI chemistry for size selection and clean up. 4. The vendor’s system must utilize specialized sealed pre-dispensed reagent cartridges for the preparation of a single library from sheared/fragmented gDNA, amplicons and cDNA starting material. 5. The vendor’s system must be a bench top solution. 6. The vendor’s system setup for preparing 1-10 libraries should not exceeds more than 10 minutes. 7. The vendor’s system must be able to process 1 to 10 libraries in less than 6 hours. 8. The vendor must be able to offer customer and technical support of the system and reagents offered. All SPRI-Based reagents and systems are supported by Beckman Coulter Inc. 9. The vendor’s system must include integrated UV decontamination. This acquisition is being conducted under FAR Part 13, Simplified Acquisition Procedures, and is exempt from the requirements of FAR Part 6, Competition Requirements. The North American Industry Classification System (NAICS) Code is 334516 and the Size Standard is 500 Employees. This notice of intent is not a request for competitive proposals. The determination by the Government not to compete the proposed purchase order based upon responses to this notice is solely within the discretion of the Government.
