THIS IS A NOTICE OF INTENT, NOT A REQUEST FOR A PROPOSAL. A SOLICITATION DOCUMENT WILL NOT BE ISSUED AND PROPOSALS WILL NOT BE REQUESTED. The National Heart, Lung, and Blood Institute (NHLBI) Office of Acquisition (OA) on behalf of the National Human Genome Research Institute, NHGRI), intends to negotiate and award a purchase order on a noncompetitive sole source basis to Invitrogen, 3175 Staley Road, Grand Island, New York 14072 to procure a genome-wide collection of Ambion Silencer Select siRNAs. The National Institute of Health (NIH) is the nation’s medical research agency and the primary Federal agency conducting and supporting making medical discoveries that improve people’s health and save lives. The National Institutes of Health has begun preparations to establish an RNAi facility to provide intramural researchers with access to the equipment and expertise necessary to conduct genome-wide RNAi screens in mammalian cells. This facility will work as a collaborative resource for doing RNAi assay development, screening, and initial follow up. Our facility will be working with investigators to develop and optimize their assays for screening, conduct the screens and related informatics analyses, then perform follow-up screens on promising leads. This purchase is for a genome-wide collection of Ambion Silencer Select siRNAs. Ambion Silencer Select siRNAs are used for the gene specific knockdown of mRNA transcripts in cells. This library has 3 independent siRNAs per gene arrayed in individual wells of 384 well plates. This item is needed to facilitate our RNAi screening program. This program is to conduct genome-wide siRNA screens in collaboration with NIH intramural investigators. This work includes RNAi assay development, screening, and initial follow up. The Ambion siRNA library will be used for genome-wide siRNA screens in human cells. Description: 1. Silencer Select HM Genome siRNA Library V4, SKU# 4397926, Quantity 1. The sole source determination is based on the fact that siRNA experiments can be hampered by poor “on-target” knockdown and undesired “off-target” effects. Ambion Silencer Select siRNAs represent a highly optimized product, incorporating the latest in siRNA design and chemistries to provide effective and specific knockdown of corresponding gene targets. The Ambion design algorithm incorporates over 90 different parameters that lead to increased efficacy and decreased off-target effects. Chemical modifications of the siRNAs also consistently enhance guide strand bias and reduce off-target effects. All of these features lead to the cleanest data possible, which is imperative for meaningful RNAi experiments. In fact, in side-by-side tests, Silencer Select reagents result in a higher percentage of expected phenotypes than those from other siRNA vendors. Ambion also offers the best guarantee in the industry, stating that 2 of 3 siRNAs will yield a better than 80% knockdown of target levels. As the genome-wide library consists of 3 siRNAs per gene, this means that multiple siRNAs targeting genes related to a particular biology under investigation will likely be identified from the screen data (i.e. a reduced rate of false negatives). No other product offers all the advantages of Ambion Silencer Select siRNAs, NHGRI have already optimized protocols for a number of on-going studies using Ambion Silencer Select siRNAs and they have proven to be reliable reagents. This will allow NHGRI to be able to extend these optimized conditions to genome-wide screening without expensive and laborious re-optimization. Only one responsible source and no other supplies or services will satisfy agency requirements. The delivery point is the National Institutes of Health (NIH), National Human Genome Research Institute, NHGRI), 9800 Medical Center Drive, Building 2B-2, Room 3005, Rockville, Maryland 20850. The acquisition is being conducted under FAR Part 13, simplified acquisition procedures, therefore the requirements of FAR Part 6 B Competitive Requirements are not applicable (FAR Part 6.001).and the resultant purchase order will include all applicable provisions and clauses in effect through the Federal Acquisition Circular (FAC) 05-43 (July 2, 2010). This notice of intent is not a request for competitive proposals. Interested parties may identify their interest and capabilities in response to this synopsis, by July 12, 2010, 7:30 a.m. Eastern Standard Time. The determination by the Government not to compete the proposed contract based upon responses to this notice is solely within the discretion of the Government. Information received will normally be considered solely for the purpose of determining whether to conduct future competitive procurement.
