Description The National Cancer Institute (NCI), Angiogenesis Core Facility (ACF), Radiation Oncology Branch (ROB) plans to procure on a sole source basis the Yeast Two Hybrid (Y2H) analysis from Myriad Pharmaceuticals, Inc., 320 Wakara Way, Salt Lake City, UT 84108. This acquisition will be processed in accordance with simplified acquisition procedures as stated in FAR Part 13.106-1(b)(1). Only one award will be made as a result of this solicitation. This will be awarded as a firm fixed price type contract. The National Cancer Institute’s (NCI) Angiogensis Core Facility (ACF) part of the Radiation Oncology Branch (ROB) has previously shown that the aryl hydrocarbon receptor repressor (AHRR) is a tumor suppressor gene in many human cancers. AHRR is a heterodimeric transcription factor and its action depends upon binding to several other factors (including ARNT and HIF). Therefore, finding the different binding partners of the AHRR is essential to understanding the molecular mechanisms underlying its tumor suppressive potential. Yeast two-hybrid (Y2H) is a molecular biology technique used to discover protein–protein interactions and protein-DNA interactions by testing for physical interactions (such as binding) between two proteins or a single protein and a DNA molecule, respectively. The ACF/ROB requires the services of a contractor to perform an Y2H experiment to globally evaluate the protein-to-protein interaction potential of the four splice forms of the AHRR gene. The contractor shall provide an analysis of the AHRR protein, or more specifically, 12 protein fragments using Y2H to determine protein-protein interactions. Myriad Pharmaceuticals, Inc. offers a unique combination of activation domain libraries, bioinformatics, industrial-scale throughput and protein-protein interaction database availability to enable a comprehensive custom yeast two-hybrid program for the discovery of novel protein-protein interactions in a minimal time frame. Myriad Pharmaceuticals Inc., has developed a database encompassing those proteins which exhibit promiscuous binding behavior. These too, are eliminated by the ProNet quality control process. ProNet is a protein- protein interaction database containing >35,000 interactions for >5400 human & >1800 mouse proteins. Thus, when any given protein is searched by Myriad, it is highly probable that the interactions from the newly searched protein will connect to interaction networks already present in the database. This allows any newly searched protein to be assigned by association to particular biochemical & cellular processes. This feature is essential in this project and Myriad is the only source with the analysis capabilities made possible by its extensive database. This is not a solicitation for competitive quotations. However, if any interested party believes they can meet the above requirement, they may submit a statement of capabilities. All information furnished must be in writing and must contain sufficient detail to allow the NCI to determine if it can meet the above unique specifications described herein. An original and one copy of the capability statement must be received in the NCI Office of Acquisitions on or before 11:00 AM EST on March 17, 2010. No electronic capability statements will be accepted (i.e. email or fax), an original and one copy must be sent to the NCI Office of Acquisitions at the address stated above. A determination by the Government not to compete this proposed contract based upon responses to this notice is solely within the discretion of the Government. Information received will be considered solely for the purpose of determining whether to conduct a competitive procurement.
