Description
The National Heart, Lung, and Blood Institute (NHLBI) Office of Acquisitions OA, DERA has a requirement for the Laboratory of Biochemistry (LBC) for the purchase of a MM-110PS microfluidizer System to replace the current Conventional cell homogenizer, such as Emulsiflex uses an external hydraulic pump to drive a piston within a larger cylinder that contains the sample. The pressurized solution is then squeezed past a needle valve. Once past the valve, the pressure drops to atmospheric pressure and generates shear forces that disrupt the cells. It is not well suited for larger volume processing. And it is prone to valve-clogging events, which we experienced a lot. Due to variations in the valve setting and seating, it is less reproducible. The M110-PS microfluidizer processors disrupt cells by forcing the media with the cells at high pressure (typically 20,000-30,000 psi) through a proprietary interaction chamber containing a narrow channel that generates the highest shear rates commercially available. The ultra-high shear rates allow for: processing of more difficult samples, fewer repeat passes to ensure optimum sample processing. Items to be purchase are; 1.Product #M-110PS, Low sample size (25 ml) benchtop Microfluidizer® processor laboratory machine for continuous high shear fluid processing, quantity one (1) each 2.Product #801.11578, Sanitary flush diaphragm pressure transducer, and digital readout manufactured with NIST traceable calibration, quantity one (1) each 3.Product #99.0450, Start-Up and Operator Training, one day start-up and training by required by factory authorized personnel. To include Travel and Per Diem, not to exceed one (1) day for completion of training. 4.Product #14.0200, 1Liter Stainless Steel Open Reservoir, quantity one (1) each. The technical specification required for the M-110PS microfluidizer System and accessories are: The Microfluidizer system must provide a highly reproducible, convenient, and efficient method for cell lysis. The fixed geometry of the interaction chamber must ensure: day-to-day reproducibility, machine-to-machine reproducibility, direct scalability from laboratory scale (20 ml to several liters) to production scale (10s of liters per minute). 1.System shall obtain minimum shear rate of 7.1 million sec-1 @ 30,000 psi operating pressure for maximum processing efficiency. Recombinant Pichia had at least 80% breakage after only 4 passes. The protein produced by Pichia will be label by 15N and 13C material, which are very expensive material. By obtaining higher percentage breakage through microfluidizer, significant saving will be achieved. 2.System shall retain product in energy zone for 3-5 nanoseconds for minimal heat absorption and, therefore, maximum bioactivity. 3.The system must have a shelf life of fifteen years or more. 4.System shall be able to obtain a flow rate of 110 ml/min on water at 30,000 psi for rapid product processing. 5.System shall process product continuously at maximum operating pressure using Electric power as the only utility from an 110V/20 amp, 230V/10 amp or 220V/12 amp plug in wall connection. 6.System shall have submersible Interaction Chamber and a 10 foot long coiled single pass heat exchanger for maximum heat removal and bioactivity in processed sample. 7.System shall utilize monolithic polycrystalline diamond in the energy zone of the Interaction Chamber for minimum wear and no product contamination. 8.System shall provide constant pressure through fixed microchannels for absolute repeatability of shear input and therefore consistent results from sample to sample. 9.System shall automatically stop processing product without exceeding set operating pressure upon accidental downstream pipeline closure for operator safety. 10.System shall maintain internal flow rates of 15 ft/sec for reliable CIP cleaning without machine disassembly. 11.System shall be designed to be chemically sterilized entire product contact path for absolute sterility control to insure operator safety. This requirement is under the North American Industry Classification System (NAICS) Analytical Laboratory Instrument Manufacturing, Size Standard is 500 employees. The Small Business Set-Aside does apply. Market research conducted by the Government has determined that there are at least two (2) small businesses available for procurement of the above item. The delivery and acceptance is F.O. B. Point Bethesda, Maryland. The Government=s estimated expected delivery date is two (2) weeks after receipt of the purchase order. The reference number NHLBI-PB-(HL)-2009-244-DDC. Proposals are due August 14, 2009 by 7:30 a.m. local time. The award will be made based upon the technical specifications of the required products and delivery period in this synopsis, and to the lowest price, technically acceptable quote meeting the Government’s requirements. The offerors must submit written descriptive literature of the product in their proposal, an itemized price list and warranties of the product quoted. The Government intends to evaluate offerors and award a contract without discussions with Offerors. Therefore, the initial offer should contain the offerors best terms from a technical and price standpoint. However, the Government reserves the right to conduct discussions if later it is determined by the Contracting Officer to be necessary. The Government may reject any or all offers, waive informalities and minor irregularities in offers received. The offerors proposals must include an itemized price lists of charges, complete description of services, complete address for invoice payment and purchase order delivery, prompt payment discount terms, the Dun & Bradstreet Number (DUNS), the taxpayer Identification Number (TIN) and certification of business size of the company. The offeror must also register in the Government’s Contractor Registry System. Note: www.ccr.gov. The proposal must reference Solicitation number@ NHLBI-PB-(HL)-2009-244-DDC.
