Description
This is a notice of intent, not a request for a proposal. A solicitation document will not be issued and proposals will not be requested. The National Institute on Drug Abuse (NIDA), Intramural Research Program (IRP) intends to solicit, negotiate and award a purchase order on a noncompetitive basis. Qualifications include: familiarity with the current projects related to the development of drugs for the treatment of cocaine abuse; intimate familiarity with the development of drugs as therapeutic agents; extensive and current working knowledge of radiotelemetry methods for the measuring of cardiovascular function in unrestrained conscious, freely moving laboratory animals; extensive knowledge of the development of parametric functions analogous to pharmacokinetic functions suitable for modeling the cardiovascular (HR and BP) response profile to drugs; familiarity with current methods for the analysis of telemetric data that provide an unbiased picture of the shape of the time course of action; and working knowledge of isobolographic analysis of drug combinations. The proposed studies will analyze the hemodynamic effects induced by drugs being developed as agonist treatments for cocaine abuse, and currently under investigation in our laboratories. These drugs, with selectivity for the dopamine transporter, have the advantage of producing behavioral effects clearly differentiated from those of cocaine. Adult (16-week-old) male rats supplied by Charles River Laboratories or equivalent will be acclimatized to standard laboratory conditions and housed in individual cages for 1 week before surgery, with free access to food and water. Rats will be anesthetized with an intraperitoneal administration of pentobarbital (30 mg/kg). A midline laparotomy will be performed, and a segment (about 1 cm below the renal arteries) of the abdominal aorta will be isolated. The catheter tip will be inserted into the aorta just above the iliac bifurcation, the transmitter will be fixed to the muscles, and the abdominal wall will be sutured. After recovery from anesthesia, the rats will be housed individually in cages placed on the receivers. Gentamicin (5 mg/kg twice daily) will be administered for 2 days after surgery to prevent infection. Blood pressure (BP) and heart rate (HR) will be recorded and analyzed by a Dataquest 4 system (Data Sciences Intl). The system consists of BP sensors (model TA11PAC40), receivers (model RA1010), a consolidation matrix (BCM100), a personal computer, and Dataquest 4 software. Hemodynamic recordings will be taken every 5 minutes, starting 2 hours before drug administration and for 24 hours thereafter. Each recording lasting 10 seconds, and the hemodynamic values of all cardiac cycles within this period (about 50 at baseline) will be averaged. The acquisition is being conducted under simplified acquisition FAR 13 and is exempt from the requirements of FAR Part 6, Competition Requirement. This notice of intent is not a request for competitive proposals. Interested parties may identify their interest and capabilities in response to this notice. The determination by the Government not to compete the proposed contract based upon responses to this notice is solely with the discretion of the Government.
