- The US Department of Agriculture, Agricultural Research Service in Aberdeen, ID has an oligo pooled assay for barley BOPA-1 will be used to interrogate 1,536 alleles in three different mapping populations. The populations have been specifically developed to map barley stripe rust resistance BCD47/Baronesse diastatic power 01Ab8219/Shellar and B-gulucan soluble fiber Falcon/Azhul. Since the USDA ARS Genotyping Laboratory in Fargo, ND already has the BOPA primers and the instrumentation Illumina BeadStation we will need to purchase the consumables for the assays. Parents and RILs will be sent to Fargo with consumables and the USDA Genotyping will run the BOPA assay. The data will then be returned to ARS Aberdeen and new molecular linkage maps will be constructed with the data. The maps will be used to identify genes controlling each of the aforementioned trails. The USDA intends on negotiating with the only known source Illumina Inc. San Diego, CA for Sentrix Universal-96 Array Matrix 1536-plex quantity five GT-103-1213 and GobadnGate Assay kit with UDG 96 samples quantity five GT-96-205. We intend on performing 96x1536 = 147,456 individual reactions simultaneously using the system developed at Illumina.
