Description
The National Cancer Institute (NCI), Nutritional Epidemiology Branch plans to procure on a sole source basis, services regarding: Urinary Sugars biomarker in the OPEN study urinary samples with Quotient BioResearch, Newmarket Road, Fordham, Cambridgeshire, CB7 5WW, United Kingdom. The supplies and services herein are being procured in accordance with the simplified acquisition procedures authorized by FAR Part 13. The North American Industry Classification System code is 541380 and the business size standard is $12M. The requirement is based on the need to generate data on the urinary sugars biomarker. The role of dietary sugars in cancer occurrence is largely inconclusive due to unreliability of self- reported intake estimates of sugars. Sugars are major contributors to energy intake; however, their intake is difficult to measure as foods high in sugars are often underreported. Recently, a new dietary biomarker has been developed to asses the validity of self-reported sugars intake. In two metabolic studies under strictly controlled conditions, we found that the amount of sucrose and fructose in 24-hour urine collections was highly correlated to total sugars intake. Results showed that 200 g of total sugars intake predicted about 100 mg of sucrose and fructose in urine. However, these findings were based on a small group of subjects; the next step is to investigate the performance of this biomarker in a larger study with free-living individuals in which dietary assessments have been validated with established biomarkers. Specific Project: An NCI-based biomarker study, the Observing Protein and Energy Nutrition (OPEN) Study, involved 484 men and women aged 40-69 years from Maryland. The OPEN study collected dietary information using two self reporting dietary instruments- a food frequency questionnaire (FFQ) and a 24-hour dietary recall (24HR). In addition, unbiased biomarkers of energy intake (doubly- labeled water) and protein intakes (24-hour urine nitrogen) were measured. The information on doubly-labeled water measurement is available form 93% of the participants and 77% of the collected 24-hour urines were verified for their completeness and analyzed for nitrogen. We propose to analyze sucrose and fructose in the urine collections and investigate how these relate to self-reported dietary sugars information as well as the doubly-labeled water and 24-hour urinary nitrogen biomarker measurements. We will also use the regression equation from the metabolic study to estimate ‘true’ intake based on the measured urinary values for sucrose and fructose. Creatinine in the urine collections will be measured to correct for urine concentration. The Contractor shall perform the following: (1) nondestructively measure fructose and sucrose in human urine samples using the sequential enzymatic color assay on the Olympus AU640 automated chemistry analyzer. (2) Deliver the data to the NCI Project Officer in a Microsoft Excel Spreadsheet. Period of Performance: Twelve (12) months from date of award. Only one (1) award will be made as a result of this solicitation. Quotient BioResearch is the only source known to the NCI that provides services measuring sucrose and fructose in urine. They are experienced with handling human samples and measuring various analytes and fulfill the NCI investigators’ requirements for study quality. The method of analyses, that QBR uses, has been validated against a reliable source Furthermore, Quotient BioResearch uses automated autoanalyser which is characterized by high precision and reproducibility and requires very small sample volume. The OPEN study is a biomarker-based study that provides two 24-hour urinary collections and measurements of two established dietary biomarkers for energy and protein, which are rather expensive and, thus, rarely done in studies with free-living individuals. This study provides a unique opportunity to investigate the performance of a novel biomarker against the performance of already established biomarkers. ^^^ This notice is not a request for competitive quotation. However, if any interested party believes it can meet the above requirement, it may submit a statement of capabilities. The statement of capabilities and any other information furnished must be in writing and must contain material in sufficient detail to allow NCI to determine if the party can perform the requirement. Capability statements must be received in the contracting office by 11:00 AM ET, on August 13, 2008.
