JUSTIFICATION FOR OTHER THAN FULL AND OPEN COMPETITION: Part I: Background information and description of requirements: The Laboratory of Tumor Immunology and Biology (LTIB) is a large translational research program with goal design, development and delivery of innovative immunotherapies for various types of human cancers. Many projects involve monitoring patient immune responses pre and post-vaccination in the vaccine clinical trials. An ELISPOT assay for interferon gamma and other cytokines production to evaluate T-cell responses was used using peptides from tumor associated antigens such as CEA, PSA, MUC-1. These assays are also used in the pre-clinical study for many investigators in LTIB. An ImmunoSpot (ELISPOT) Reader is required for the measurement of the number, the size, and the intensity of the cytokine producing specific T cells. The current ELISPOT Reader in LTIB is a first generation ELISPOT Reader. This ELISPOT Reader has limited functions and applications. In addition, the current ELISPOT reader gives diffused images and extra effort is required to distinguish the different between dirt from small spots in some samples. An advance model ELISPOT Reader is essential for the precise determination of patient immune responses pre vs. post vaccination.
