Description
The National Cancer Institute (NCI), Division of Cancer Epidemiology and Genetics (DCEG), Nutritional Epidemiology Branch (NEB) plans to enter into a sole source basis procurement in accordance with 13.106-1(b)(1), with Guillermo Perez-Perez Laboratory, New York University Medical Center, 550 First Avenue, New York, NY 10016 for laboratory assays to measure antibodies to H. pylori Whole Cell and CagA antigen on up to 640 PLCO stored serum samples. Antibodies will be measured to H. pylori Whole Cell and CagA antigen on up to 640 PLCO stored serum samples. Serum will be evaluated for IgG antibodies to whole-cell (WC) by enzyme-linked immunosorbent assay (ELISA). Serum samples will be assayed in duplicate, and the results expressed as optical density ratios relative to simultaneously analyzed laboratory standards. An antigen-specific ELISA will be used to determine serum levels of IgG antibodies to H. pylori using a preparation of sonicated whole cells of five clinical H. pylori isolates. An absorbance index (AI) will be calculated from mean absorbance at 405 nm of two assays per sample, in relation to reference sera. A previously validated cutoff point of AI > 1.0 will be used to define H. pylori positivity. When two assayed aliquots provided indeterminate results (i.e., the values straddle the seropositivity threshold), additional aliquots will be analyzed and the average of all results will be used to determine serologic status. CagA status will be determined by an ELISA based on the presence of serum IgG antibodies against orv220, a 65,000 Dalton recombinant cagA-encoded protein purified from Escherichia coli.. CagA positivity (representing carriage of a cagA+ strain) will be defined as an optical density > 0.35, as validated previously. The samples will be stored at -70 degrees Centigrade. Period of performance shall be for one year from date of award. Guillermo Perez-Perez Laboratory, New York University Medical Center is the only source known to the NCI Measure antibodies to H. pylori Whole Cell and CagA antigen on up to 640 PLCO stored serum samples which will meet the research needs of the project in a manner consistent with prior studies. To maintain continuity in on-going research projects and avoid introducing new variables into the project, it is of critical importance that the procedures be conducted in the same laboratory so that there is no variation or interpretation of the results. Assuring no intra-laboratory variation is essential to maintain validity of the overall evaluation. This notice is not a request for competitive quotation. However, if any interested party believes it can perform the requirement as detailed they may submit a statement of capabilities. The statement of capabilities and any other furnished information must be in writing and must contain material in sufficient detail to allow the NCI researcher to determine if the party can perform this requirement. An original and one copy of the capability statement must be received in the contracting office by 1:00 pm EDT, on August 31, 2007. Faxed and emailed capability statements are NOT authorized. A determination by the Government not to compete this proposed contract based upon responses to this notice is solely within the discretion of the Government.
