Description
The National Cancer Institute (NCI) Division of Cancer Epidemiology and Genetics (DCEG), Occupational and Environmental Epidemiology Branch (OEEB) plans to procure on a sole source basis with the University of Milan, Department of Occupational and Environmental Health, Milan, Italy to investigate the role of telomere length and methylation status, including global methylation and methylation in E-cadherin, p16, iNOS, TLR-2, and hMLH1 genes, in DNA from blood or buccal cells in relation to gastric cancer risk in a nested case-control study within the Shanghai Women?s Health Study. The contractor shall: 1) Perform PCR-based analysis of telomere length on 525 blood or buccal DNA (henceforth referred to as DNA) samples in duplicate from the Shanghai Women?s Health Study; 2) Perform bisulfite treatment priori to methylation analysis for 1050 DNA samples; and 3) Perform PCR amplification and pyrosequencing for global methylation analysis and methylation analyses in the promoter area of E-cadherin, p16, iNOS, TLR-2, and hMLH1 genes for 525 samples. The Molecular Epidemiology Laboratory of the University of Milan, Italy, is a leading molecular epidemiology laboratory that has acquired the expertise and resources critical for conducting the proposed analysis in large environmental epidemiologic studies. The University of Milan is the only institution worldwide to have pyrosequencing equipment for methylation analysis dedicated exclusively to environmental health-related research. The pyrosequencing technology on bisulfite treated DNA has the sensitivity and accuracy to detect very small changes in DNA methylation, such as those that are of interest in epidemiology studies. The laboratory implements standardized quality control procedures that take full advantage of the pyrosequencing potential. In particular, the QC procedures permit accurate determination whether bisulfite treatment, a critical step in DNA methylation analysis, resulted in complete cytosine conversion. The real-time PCR assay that the laboratory uses for telomere length analysis also have the high sensitivity and precision needed to test the study hypothesis. The University of Milan conducted analysis of DNA methylation in relation to gastric cancer risk for the Division of Cancer Epidemiology and Genetics, NCI, under purchase order MQ606468. The conduction of the DNA methylation study in the same lab will allow NCI to simplify the control of laboratory operations and guarantee standardized quality control procedures. This will enhance the comparability of results should NCI decides to conduct pooled analysis for more detailed evaluation of gene-environment interactions in the future. This acquisition will be processed under FAR Part 12 - Acquisition for Commercial Items and will be made pursuant to the authority in FAR 13.5 to use simplified procedures for commercial acquisitions. This is not a solicitation for competitive quotations. However, if any interested party believes it can provide the above service they may submit a statement of capabilities. The statement of capabilities and any other information furnished must be in writing and must contain material in sufficient detail to allow NCI to determine if the party can perform the requirement. An original and one copy of the capability statements must be received in the contracting office by 1:00 p.m. EDT, on August 23, 2007. Faxed and emailed capability statements are NOT authorized. A determination by the Government not to compete this proposed requirement based upon responses to this notice is solely within the discretion of the Government.
