Description
The National Institute of Mental Health (NIMH) is seeking information on all sources capable of providing screening and testing services to continue the NIMH Psychoactive Drug Screening Program (PDSP), which was first initiated in 1992. To be considered responsive to this request for sources, the organization must be qualified to: (1) establish and validate assays for cloned human or rodent receptors by cloning a receptor, maintaining a stably transfected cell line expressing the receptor, and establishing the binding and functional assays for the receptor in accordance with standards established in the published scientific literature; (2) conduct high-throughput screening of compounds in broad-based primary and secondary pharmacological assays (radioligand binding and enzyme assays), functional assays, and assays to assess brain penetrance, bioavailability, and cardiac toxicity (Note - the screening services are intended primarily as a screen for compounds that have previously been shown to possess pharmacological, biochemical, or behavioral activities relevant to NIMH, rather than for the purpose of large-scale, random screening of natural products or combinatorial libraries) (3) provide an electronic data file of screening results for each compound suitable for inclusion in a scientific publication; and (4) update and maintain a Ki database for compounds and drugs. Qualified sources must be able to receive and test approximately 2,500 (but could range from 2,000-3,000) coded samples (synthetic compounds, small molecules, gene products, and natural product extracts) per year, in a wide variety of human and rodent CNS receptor and enzyme screening assays and functional assays, and to provide an electronic data file for each of the screened compounds. (A list of targets of interest to NIMH can be found at http://pdsp.med.unc.edu/pdspw/clones.php.) At least 85% of these samples are to be tested in human recombinant receptor assays. Samples for screening can include, but are not limited to, novel chemical entities, structural analogs of lead compounds, genes or gene products, small molecules, enzyme inhibitors, and natural products. Based on historical data, the projected annual assay workload for this requirement is estimated at: 1) 15,000 primary pharmacological assays; 2) 1500 secondary pharmacological assays; 3) 15,000 functional assays; 4) 1,000 brain penetrance, bioavailability, or toxicity assays; and 5) 5-10 new assays developed and validated. These are estimates, which can fluctuate as much as 50% higher or lower than the number of assays stated. The NIMH Psychoactive Drug Screening Program currently operates under a Determination of Exceptional Circumstances (DEC), which allows a deviation from the standard Patent Rights clause in FAR 52.227-14, to ensure the confidentiality and protect the intellectual property and patent rights of an investigator or party supplying a proprietary sample or composition of matter for testing through the program. It is the intention of the Government to seek a continuation of that DEC to allow use of a similar deviation in the currently proposed requirement. Any business or organization that believes it has the capability to fulfill this requirement should submit a brief narrative response (no more than 5-10 pages, single-spaced), indicating previous experience and capabilities related to the described work. Capability Statements should clearly state/describe: 1) your experience and qualifications to perform this work; 2) your type of business or organization, including: (a) size (number of employees); (b) management structure and history; (c) eligibility under 8(a) or other set-aside programs, as applicable, for NAICS Code 541710 (500 employees); and 3) any specific resources, equipment, available to perform this work.
