Description
The Centers for Disease Control and Prevention intends to issue a sole source purchase order to Nimblegen Systems, Inc., 1 Science Court, Madison, WI 53711, for high-resolution microarray-based subtyping of Clostridium botulinum. The vendor utilizes maskless array synthesis technology in order to generate microarrays with large probes (up to 70-mer) in a high density format (up to 390,000 features). The technology also allows the vendor to synthesize new array design within a day. The Botulism Public Health Research and Preparedness Unit (BPHRPU) is currently investigating the genotypic diversity among C. botulinum strains in their culture collection in order to develop improved subtyping methods. Strains will be compared within the collection with that of the genome sequenced strain, C. botulinum Hall type A strain using a comparative genome hybridization technique followed by re-sequencing arrays of regions that permit discrimination among strains of C. botulinum. The vendor will be required to develop the microarray using 70-mer probes titled approximately every 20 base pairs. The entire ~4 megabase pair C. botulinum Hall type A strain will be represented on a single microarray to achieve ?? 90% coverage. Synthesized arrays will then be shipped to CDC for hybridization with genomic DNA and data derived from these experiments will be used to generate re-sequcncing arrays which will enable all the DNA sequencing of up to 48,000 bases to be determined from a single array. The vendor will also be required to design and synthesize the microarrays and provide technical/bioinformatics assistance. BPHRPU requires the ability to change the chip design during the study. To allow this flexibility in chip design, it is required that maskless array synthesis technology be used to synthesize the microarrays. Nimblegen was the only company identified that utilizes maskless array synthesis technology and therefore has the flexibility to change the array design within 24 hours. In addition, technology developed by Nimblegen will permit BPHRPU to design and generate resequencing microarrays of specific regions of genomic DNA that may provide useful subtyping information.CDC believes that this requirement is met by only one provider. This procurement will be processed under the authority of FAR 6.302-1 and 6.302-2. Only one responsible source and no other sources will satisfy agency requirements. No solicitation is being issued.
