" THIS IS NOT A SOLICITATION ANNOUNCEMENT. THIS IS A SOURCES SOUGHT SYNOPSIS ONLY. THE PURPOSE OF THIS SOURCES SOUGHT SYNOPSIS IS TO GAIN KNOWLEDGE OF POTENTIAL QUALIFIED SOURCES FOR PLANNING PURPOSES ONLY FOR THE U.S. ARMY MEDICAL RESEARCH ACQUISI TION ACTIVITY (USAMRAA), FORT DETRICK, MARYLAND. USAMRAA is seeking a Contract Research Organization (CRO) to perform the attached Statement of Work. This announcement is not a formal solicitation and is not a request for proposal. No contract will be a warded from this announcement. No reimbursement will be made of any costs to provide information in response to this announcement or any follow up information requests. Availability of any formal solicitation will be announced under a separate announceme nt. This synopsis is for information and planning purposes only and is not to be construed as a commitment by the Government. Each response must reference the sources sought title. All proprietary information should be marked as such. Responses will be held confidential, if requested. Statement of Work Evaluation of the acute and persistant/resting memory phase responses of rabbits to rPA vaccine 1. Purchase 16 New Zealand white rabbits, 8 males and 8 females, at 2.7-3.1 kg each. Allow animals to acclimatize for 1 week, if necessary. 2. Prebleed 16 rabbits prior to the start of the experiment for (i) 1 ml of serum and (ii) 14 ml of blood to isolate PBMCs. These cells will serve as baseline values and negative controls for the lymphocyte proliferation assay, CD4/CD8/CD3 FACS, and real -time RT-PCR. 3. One week after the pre-bleed, inject 50 ?g rPA/Alhydrogel vaccine intramuscularly (0.5 ml each) into 12 rabbits (6 male and 6 female). Inject 4 rabbits (2 male and 2 female) with PBS/Alhydrogel (age-matched controls). Vaccine will be supplied by USAM RIID. 4. Divide the rabbits into two groups A and B each of 6 rPA-immunized rabbits (3 males and 3 females) and 2 controls (1 male and 1 female). 5a. Group A (acute memory phase); A. Bleed 8 rabbits on days 2, 4, and 6 for 2 ml of blood. Send frozen serum to USAMRIID. B. Sacrifice 8 rabbits on day 7 by exanguination and harvest and purify PBMCs. a. Lymphocyte proliferation assay using cultured PBMCs stimulated with PA. Include positive and negative controls. USAMRIID will provide PA. b. Real time RT-PCR for IFN-? and IL-4 RNA using the PBMCs. Include reference standard (i.e. glyceraldehyde-3-phosphate dehydrogenase (GAPDH) or ?-actin). Published GenBank rabbit sequences for IFN? accession number AB010386, for IL-4 accession number AF 169169, for GAPDH, accession number NM001082253, and for ?-actin, accession number AF309819. (Please describe the detection chemistries, instrumentation, and quantification method to be used). c. FACS on cultured and rPA-stimulated PBMCs to identify/separate CD4/CD8/CD3 T cells. Include positive and negative controls. USAMRIID will provide PA. 5b. Group B; Bleed 8 rabbits on days 2, 4, 6, 8, 10, 12, 14, 16, 18, and 21 for 2 ml of blood. Send frozen serum to USAMRIID. The blood volume of 2.7-3.1 kg rabbits is about 56 ml/kg or about 150 ml at a minimum. It is safe to remove 7.5% blood on a w eekly basis (11 ml). Thus, each week the volume would be 6-8 ml, within the accepted guidelines. 6. Group B; On day 21, boost im 6 rabbits with rPA/Alhydrogel and 2 rabbits with PBS/Alhydrogel as used in the primary injection. 7. Group B; Bleed 8 rabbits on days 2, 4, 6, 8, 10, 12, 14, 16, and 18 after the booster injection for 2 ml of blood. Send frozen serum to USAMRIID. 8. On day 98 (77 days after the booster injections) for each of 8 rabbits, A. Collect 3 ml of blood in a serum tube. Send frozen serum to USAMRIID. B.. Sacrifice rabbits by exanguination and harvest and purify PBMCs. a. Lymphocyte proliferation assay using cultured PBMCs stimulated with PA. Include positive and negative controls. USAMRIID will provide PA. b. Real time RT-PCR for IFN-? and IL-4 RNA using the PBMCs. Include reference standard (i.e. glyceraldehyde-3-phosphate dehydrogenase (GAPDH) or ?-actin). Per diem for 8 rabbits 21 days and 8 rabbits for 112 days. 10. Serum can be sent in two shipments; after the final bleed of the boosted animals (day 39) and after the final sacrifice bleed (day 98).
