Description
The Department of Agriculture (USDA), Animal and Plant Health Inspection Service (APHIS), Veterinary Services (VS), Center for Veterinary Biologics Laboratory in Ames, Iowa intends to acquire Tuberculin, PPD Bovis, Intradermic in 10 ml vials. Veterinary Services requires an estimated 200,000 ml vials of PPD Bovis per year. Additional 20% optional quantity for each 4-1 year option (20,000 ml vials). The PPD Bovis Tuberculin is required as a diagnostic test reagent in the National Eradication Program for bovine tuberculosis. It is a product derived from the culture of the AN-5 strain of Mycobacterium bovis in a modified Reid?s media for 8-12 weeks in constant temperature incubator equipped with a device to increase the relative humidity. These cultures are grown as floating cultures to maximize the yield of tuberculin. The 8-12 week culture is heat killed with steam under pressure at degrees C for 30 minutes. The culture media is filtered, preserved in a 5 percent phenol solution, concentrated to 2 percent of the original volume, and precipitated in ammonium sulfate to yield a product with a protein concentration of 1 mg per ml. Each serial lot of tuberculin shall be tested for purity, safety, potency, and special chemical characteristics in accordance with the conditions presented in Title 9, Code of Federal Regulations (9CFR) Parts 113.203 113.3 or in Section V of the approved outline of production, which is on file with USDA, APHIS, VS.
