Description
The US Department of Agriculture, Agricultural Research Service in Riverside, CA has a requirement for a Real-Time PCR System with the following characteristics, Detector: Scanning photomultiplier tube (PMT). PMTs for multiplex PCR, Capability of multiplex detection of four dyes simultaneously at wavelengths that provide minimal overlap of excitation and emission bands, resulting in minimal crosstalk between dyes used. Capable of using most commercially available dyes, including FAM, SYBR Green, TET, HEX, JOE, VIC,TAMRA, Texas Red, ROX, Cy5, Cy3, ALEXA Fluor 350, and multiple detection chemistries, molecular beacons and hydrolysis or Taqman probes. We will need to use the same dyes with fluorescence microscopy for in situ analysis of gene expression in cells. Format: 96.well plate, Reaction volumes: 10- 50 ul; optimized for 25 ul, Excitation range: 350 -750 nm Emission range: 350 -700 nm, Dynamic range: 10 orders of magnitude, molecular beacon designer probe and primer design software and software for quantification analysis for gene expression experiments as well as capability of calculation of standard curve corrections notebook computer with software allowing real-time monitoring of reactions, melting curve, analysis, automatic calculation of melting temperatures from dissociation experiments.embedded computer to safeguard data in the event of power loss or communication failure to the external computer.5-position, dual customizable filter wheels (excitation and emission) with filters to be chosen by research leader based on which dyes will be used.Temperature range: 25 -95 C or wider Temperature variation across 96-well plate: +/- 0.25C Rapid ramp time: Up to 2.5C per second, Baseline correction individually optimized for each data set. Instrument must include a PCR license, training to include detailed instructions on hardware and software use, optimizing conditions,trouble-shooting, and designing and optimizing multiplex PCR.
