Description
CORRECT TITLE TO: LOW COPY PLASMID (BAC-END) SEQUENCING. Provide bi-directional sequencing of 7,680 clones, using the following methods: colony picking of inoculation of clones from 96 or 384 glycerol plates, cell culturing and DNA purification using SPRI or equivalent, dye-terminator fluorescent sequencing reaction set-up, sequencing product purification using CleanSEQ or equivalent, detection of sequencing fragments via capillary electrophoresis, creation of chromotograms, PHRED quality scores, and FASTA sequence files, and data transfer via secured FTP site. A test sample will be required prior to award of the entire sequence. The test tample will be 96 clones, bi-directionally, for a toal of 192 sequencing reads. The test sample shall be completed within seven calendar days of award. The entire sequence shall be completed within 14 calendar following receipt of Government materials. Sequence quality shall be85% total sequences with PHRED QV greater than or equal to 20 for more than 500 bases. Sequences shall be named according to a universal naming structure, with components having been pre-determined by the USDA, Agricultural Research Service, Animal Disease Research Unit, in Pullman, WA. The offeror shall provide example of past experience on same or similar work in order to demonstrate your ability to perform this work satisfactorially and in accordance with the specifications. Provide description of prior projects, date of contract, dollar value, period of performance (duration) and reference name and phone number for the project. Provide at least 3 examples.
