Description

The USDA, Agricultural Research Service in Albany, California has a requirement for the design and synthesis of 70-mer Oligonucleotide probes for microarray experiments. Must provide full bioinformatics service to design 10,000-20,000 70-mer oligonucleotides from private or public assembly of wheat expressed sequence tags (EST). Designed from 750-1000 bp from the 3? end of available gene; Tm normalized and designed to minimize homopolymeric stretches, highly repetitive regions, low-complexity regions, and secondary structure. The sequence must be optimized by BLAST against all known open reading frames (ORF) to minimize cross-hybridization. The synthesis must be 70-mer oligonucleotide probes; quality control by OD and batch PAGE or CE analysis; and provided as salt-free, concentration normalized and lyophilized in 96-well plate format. Must provide free aliquoting of probes into 3-4 daughter plates; 200 nM synthesis scale with 16-18 nmole guaranteed yield; and provide a 96-well test plate of oligos for printing and hybridization test. Delivery must be two weeks from receipt of oligo order.
