Description
USDA, ARS, SAA intends to solicit and negotiate with only one source, Telechem International, Inc/Arrayit to do gene expression profiling using known pig genes as probes in order to produce 450 70-mer oligonucleotides printed on optically flat superaldehyde microarrays substrates. 150 microarrays will be printed immediately from oligos in storage at their facility with at least 300 printed at a later date as needed. Specifications of this process are: USDA provides pig EST?s and tentative consensus sequences from a pig gene library, 450 70-mer oligos are to be designed from these sequences and must be specific for each gene. This directive ensures that the oligos are mapped to within 1000 nucleotides of the 3' end of the selected genes to eliminate poor hydridization due to inefficiencies in cDNA probe synthesis; designed oligos are selected to avoid sequence repeats and long stretches of polyA,G,C and T, also the melting temperatures of the oligos are matched within a selected range; and each oligo is checked against gene sequence databases to avoid cross-reactivity with transcript species of <70% sequence identity. Oligos must be synthesized and printed in triplicate on optically flat glass. A control set of E.coli genes (10) will be included on the arrays to be used as negative or positive controls. Aliquots of poly A RNA corresponding to these genes must be provided. The company must have the facilities and capability to store for a period of up to 2 years and facilitate microarray manufacturing as needed. Interested parties should submit written responses including prices and technical data sufficient to determine capability to meet the entire requirement and must be received in this office within 15 calendar days after publication of this notice. No telephone inquiries will be accepted. It should be noted that no contract will be awarded on the basis of offers received in response to this notice, but will be used to determine if a comparable source more advantageous to the Government is available. If no affirmative written responses are received, negotiations will be held with TeleChem Internation, Inc/Arrayit without further notice. This acquisition is being conducted under the Federal Acquisition Streamlining Act, Simplified Acquisition Procedures. The anticipated award date is July 14, 2003.
